My personal collection:
Little King's Story - Very overlooked game. It's an action/RTS hybrid with tons of charm, and although it sort of looks like animal crossing, it is very challenging and satisfying. It was developed by something of a supergroup consisting of devs who were behind decent titles like the original Harvest Moon games and Hotel Dusk.
Zack & Wiki - Essentially a point and click adventure for the Wii by Capcom. If I were to compare it to a specific game, it would probably be Sierra's Gobliiins, where the focus is more on getting through a stage filled with puzzles than story-telling. The puzzles and scenarios in this game are very clever and challenging. I simply love this game.
Punch-Out!! - If you liked the Punch-Out! series growing up, you owe it to yourself to at least rent this game. It's like a love-letter to fans of the NES game, in particular. It was unfairly criticized by some for being short and lacking in challenge, as the game's first mode, "challenge mode" is fairly easy; however, that's really only the first half of the game. When you play through title defense mode, where Little Mac has the belt and must defend it, every single character is completely revamped from head to toe, and it's a hugely different experience. Prepare to be KOd by an armored, more unorthodox Glass Joe.
Smash Bros. Brawl - Doesn't really require an explanation. If you like any of Nintendo's franchises, you'll dig this game for its fan service. I do feel it's the best in the series.
A Boy and His Blob, which I no longer own, was pretty good, but I got bored of it half way through. A lot of people were expecting this to be a downloadable wii-ware title, and I do wonder if the game would've sold better if it were a little leaner (didn't need 80 stages, in my opinion), content-wise, and had been released that way.

Super Smash Bros. Brawl and the Metroid Prime series are definitely among the best games for the Wii.

Would rent both first though, it isn't for everybody. I can see Smash Bro's being good at what it does but it is not my thing. The 3d-Metroid games aren't that easy to pin down either, you might hate them.

Seconding Red Steel; it is simply an amazing game, musics in particular are nice. Also seconding about the "not everyone likes Metroid"; for some weird reason I pretty much hated the Corruption (haven't played the others).
If you haven't played the original Broken Sword, and like adventure games, the new Directors edition, I felt, is a must-have. There are some small annoyances like that the voice-acting hasn't been remastered for the French language settings etc., but all-in-all it is solid gold.
Another game I would like to recommend which hasn't been mentioned yet is, in case you are interested in soccer (who isn't?), Pro Evolution Soccer. It handles very differently compared to the other platforms it has been released on.
Yet another game I would like to recommend is The Godfather Blackhand Edition. I got it for a present a longish while back and can't remember when I would have been more positively surprised about a game. I didn't have any expectations and hadn't heard about it at all and it just sucked me in completely. Granted, I'm a sucker for the Godfather -movies (again, who isn't?).

My brother owns a wii and we've been enjoying it. This is the only console we've owned since a Turbo grafx 20 years ago. Here are the games that we've really enjoyed:
Boom Blox: this was a fun one 2-player, except we hated how you have to unlock everything. The problem is, some of the game modes aren't fun at all and we wanted to just skip them, but couldn't. Good production values, a fun, cute little game, although I don't know if I'd play it single-player.
Mario Galaxy: I didn't play this one but my bro did and he really liked it. As he described it, there's tons of fun and interesting and unusual worlds with lots of variety. It's a very interesting and colorful 3d platformer. Have a look to see if it's up your alley.
Resident Evil 4: Both of us really enjoyed this one. Being primarily a PC gamer it was hard for me to get over the extremely low resolution and some of the controls, but aiming with the stick works pretty well (although, I think the Metroid Prime games did this the best on the wii). After a while you get used to the resolution, and it's actually a very fun game, and pretty long, too. I like how when I played it, I upgraded my guns completely differently from my brother (Red9 all the way!).
Gamecube Resident Evil remake: ironically my favorite game for the wii was the Resident Evil remake for the gamecube. The controls were really weird at first but I REALLY liked this game. Except for the one time when I ran out of save ribbons and had to go like 45 minutes and fight 3 bosses until I found more. I just loved the tension plus the graphics are stunning! Currently I'm playing through Resident Evil Zero and while it's pretty good and I'm enjoying it, it's just not as good as the remake.
Lost Winds and Lost Winds 2: wiiware sidescrollers where you walk around beautiful hills and caves controlling the wind to advance. They're really short, but very peaceful, nice, and well-made games that probably anybody can enjoy.
Viewtiful Joe: another gamecube game but probably my favorite sidescroller (after Cave Story of course). The graphic style is wonderful (screenshots don't do it justice, you need to see it in action), it's funny, and Joe's powers are satisfyingly effective.

edit: I forgot to mention Zack and Wiki! This is a really fun puzzle-y game that my brother and I worked on together. I didn't like the dialog, it seems geared towards 2 year olds, but the puzzles are good (some are pretty difficult!) and some of the locations are really neat.

Everyone's already beat me to most of the recommendations, so I'll just second a few opinions.
Metroid Prime Trilogy: While I haven't played this rev yet, I have played all the Metroid Primes. MP1 and 2 redone with the tight controls of MP3. You can't ask for any more.
Zack and Wiki: Fun. As. Hell. Simple as that. You can find a used copy on Amazon for $20. Geddit. :mad:
Resident Evil 4: Take the graphics of the Gamecube version, add in all the extras from the PS2 version, mix well with awesome Wiimote controls. The end result is a great damn game.
Super Mario Galaxy: We all love Mario. But we all love Mario with crazy gravity even moreso. Great game, though it ranks slightly behind...
Super Mario 64 (VC): One of the greatest games ever made in my humble opinion. Graphically, it hasn't aged well, but it's so fun you won't care in the least.
Zelda Ocarina of Time & Majora's Mask (VC): Super Mario 64 and these two games justified the Nintendo 64. No Zelda before or since have matched these two for pure unadulterated adventuring fun...though Windwaker does comes close.
Mercury Meltdown Revolution: Great puzzler with good Wii tilt controls. Will drive you insane, but in a good way.
Any one of the above mentioned will keep you entertained for hours on end. Have fun. :D
