I don't know how it got changed, but I can definitely vouch for it causing problems, I just never thought to suggest it might be the problem. One of my testers for DCE had it set to 4 or 8 and experienced all manner of inexplicable issues with a mission which is mostly camvator sequences, and which has many sounds going on simultaneously between the voice lines, music and sound effects. When he set it to 12, it all went away. I've already got a warning in my readme to warn users to check their Audio channels setting before starting the campaign and make sure it's at least 12 and preferably higher. I don't know if this is a leftover from hardware and system limitations of the days when Thief was released and we can safely go much higher these days, or if you need to check your sound card or driver capabilities before setting it much higher than 12. Glad you figured it out.

I was going to suggest audio channels but I'm glad you found it. One way to verify that your mission is not the problem is to load another mission and see if you experience the same results.

Yeah I used to mess around a lot with DromEd, almost resulting in crashes every time so I always have a backup version somewhere, I'm not even surprised when something isn't working ^^ DromEd'll be the death of me. Well I'd better rush the level before another thing goes wrong ! Thanks guys :)
