What the title said. Any particular moment that while playing really stuck out in your mind- for good or bad. It might be something that defines the game, or something that was just incredibly well-put together so you're left sitting there going "damn!"
Moments. I knows you has them. Gimme.
To get things started:
1) Half-Life 2: Episode 2: The seige at the inn. With the music (Eon Trap, for the curious), it's the defining moment of the entire HL2 series, for me, with the underside of the train bridge in HL2 and the zombie rampage in E1's hospital being close seconds.
2) Thief 2: Standing on the ledge on the side of Angelwatch, realizing that this really feels like something out of cyberpunk, what with the semi-futuristic bent of Angelwatch itself and the tall, tall, tall buildings.
3) Deus Ex: The Helios ending. 'Nuff said.
4) MGS3: The fight with The End.
5) Fallout 1: The broken vault in Necropolis, and the Glow in its entirety.

Getting the fire arrows. Firing an arrow into the sun as it rose over Lake Hyrule was just one of those moments.

A hammerite standing in a haunted cathedral, wtf? Anyway, sup head meet blackjack. That turned out to be quite a shock.

Moments that have stuck with me? Pfft, hundreds.
-getting to the driving level in Vendetta (C64) and realising just how varied the gameplay could be for one game
-playing Sonic the Hedehog 2 for the first time on my friends Sega Mega Drive
-playing the Wolfenstein 3D demo for the first time on another friends PC
-playing the Duke Nukem 3D demo, getting to the poolhall and noticing that you could actually PLAY TEH POOL. game within game. meta.
-playing the Quake demo for the first time and realizing that the character actually leaned slightly to the side when he strafed. omg. groundbreaking.
-jumping at a wall for 10 minutes before figuring out the mantling in the Thief 2 demo
-shooting a chef in the back of the head in Hitman and seeing him getting flung forward over the sink and then slowly slide backwards and colapse onto the floor thanks to the awesome ragdoll physics.
-coming home to your family in the first level of Max Payne
-the first sunrise in GTA3
...to name a few.

I almost had a heart attack at the start of that mission and died about 10 times just trying to avoid that cruiser.
Here are my top five moments
1) Pretty much all of the cradle from TDS, that whole mission is so awesome and eerie.
2) The intro to Soul Reaver, that was possibly one of the most awesome FMV's and cutscene that I've ever seen
3)The Psycho Mantis moments in Metal Gear Solid 1, he was one of my most memorable bosses as well. I love some of the things he says before his battle like "What?You don't like girls?"
4)KOTOR1- When you find out
that you are Revan and equally KOTOR2 when you fight Darth Nihilus
5)Vampire:The Masquerade-Bloodlines, a couple of moments in this one: when
you find out that Jeannette and Therese are the same person, The Ocean House Hotel and meeting Pisha in the abandoned hospital.

-Seeing the guard in power armour for the first time at the BoS in FO1. I froze there for a few minutes, too terrified to approach.
-Jumping into a canyon, do a front-roll, then stabbing a Reborn to death in one swift movement in one of the later levels in Jedi Academy.
-My first MP experience in NWN1. The group was excellent, everyone stayed in character, there was no canned responses, and the DM was very flexible and can change the story on the fly. Sadly, that was also one of my last MP game, partly because I never had another game quite like that and quickly lost interest.
-Gorion doing the Magic Missiles Spam.
-Seeing the Shadow Dragon in BG2. That was the first time I saw a creature that big in any game.
