1- I don't remember how much loot I put in the mission. Is there a way to know it?
2- Maybe I made a mistake. I gave belt pouches to some guards, but I distributed them depending of difficulty. I.e. there are 2 guards patrolling and each of them has a pouch, but one of the pouches will only appear on 'easy' and the other only on hard and expert. There is even a guard with pouch which will only appear in expert mode.
The question is: will the game take in account the total loot present in the mission, as I see it in Dromed, or it will be properly distributed depending of the difficulty?

I'm sure there's a more elegant way, but the basic way is to choose "Generate Report" from the "Tools" menu in DromEd. Just click OK (the default settings are fine) and open the file "default.rep" in your Thief directory. At one point in the file, there will be a line starting with "Total loot:" that gives you the loot values divided into gold, gems, goods, and specials. The other reports in that file can also be very useful.

The total loot statistic will be based on the difficulty. As long as the pouches are actually destroyed on other difficulties, instead of just hidden away unattached, they won't count towards the total.

Just start the game, finish it by pressing CTRL-ALT-SHIFT-END and read the stats screen. That's all. Repeat two times more for other diff. levels.

Right. This will give you always the right answer.
Relying on DromED reports is not as safe, as one might think.
Over the last 2 years I came across a couple of mission, where authors had added loot in such way to the mission
that some loot would not even show up in the DromED loot report, nor LazyTaffers lootlist tool.
Don't know what 'fancy' method these authors had used (probably via scripts ?), but as long as you stick to the 'traditional methods' to add loot to your mission, the reports should be precise as well.

http://www.ttlg.com/forums/showthread.php?t=131030) The Trickster's Loot Locator tool would then produce a handy lootmap for you then, including info which item is on which AI on which difficulties, in case you need it.

This reminds me...
In "The Mirror" FM I used one random piece of loot. It can appear or not, depends on player's actions, so the final result (loot total) may vary.
But if an author use such a thing, he (or she) will never forget about it!
