Gloria, where the sewer is, you'll get the key for that later, right before the steps, to your right, is the vine you climb. Took me a bit to find it too.

Gloria - the poisoned one? Frob the original bottle onto it & the poisoned one should turn up in your inventory

I'm not sure which wine bottle you mean. First, you have to get the wine bottle from the chest in the wine cellar which is under the streets near the inns. Then you have to "use" that bottle on the one on the mantle in Rynolf's room. In other words, you have to swap them. The same is true for the spice bags.
