Well, he doesn't respond to that at all, so no luck there. Might be a difficulty difference, i'm playing on Hotshot difficulty.
The lab is in the same building just opposite to the kitchen and the
explosive charge is on the top shelf in the back.

I'm lost too. I can't get out of the suburbs!
[SPOILER]I visited the keysmith, found out I needed 500 gold to get the key (I assume the Old Gate is the only viable path to Downtown). But I can't get 500 gold for the life of me! I have 450, and I know where I can find another 10 -- though jumping in the water in the building to get the coin on the grating busts my ghost every time... the maintenance technician always hears me and alerts.
Still, that would only bring me to 460. I think I've been in every building in the suburbs that is accessible; I even picked the purse of the guard at the Main Gate. I found the secret door in the Library (where the clue about ice is). I've swum the river from one barred end to the other; I've been all through the sewers (I think!). I got the Post-Office coin and the Library inkstand; I can get into the PO Storage area, but I have no idea how to open the box where something or other is, or even what box that would be (there is one small box that is turned funny; maybe that's it -- but it doesn't frob). But I just can't get enough $$ to buy the key.
I read that there was a gem in the lamp in the Andromeda, but I don't know what or where that building is.
I have the objective to find Athropos, to get the 500 for the key, the no-kill, and of course the first one to snuff out the candle.[/SPOILER]
What should I do next?

To all players: to avoid misunderstanding each other please write in your posts what dif. level you are playing on. There are big differences between them.
For example, Poison_the_well, you can just TAKE piece of ice from the Ice Keeper, on any other dif. level you have to GET it.
The Elusive One, the sequence of your actions must be logical a bit. Get the objective to kill first, THEN try to kill. Otherwise some things won't work. You can't blow the statue before you are really need to do this.
Dafydd, I don't know exactly where you have already been, but there is (are?) 800 of gold in Suburbs. Looks like you haven't guess to do some tricks.

The first question is: have you got the semi-versatile key from the Keymaker? Just take it on the shelf. You can gather 480 loot without this key, not more.
The wall water pipe you can see nearby the South bridge is climbable. That'll give you some loot;
you have to get rope arrows from the Weapon shop in Silver Hills;
use them in south-east part of Dark Side. Search carefully above you in the area around the passage nearby the torch. When you reach the roof, you'll be able to jump to the nearby balcony or to continue your roof exploring;
the pub in Dark Side has the entrance from the sewer.

Yeah, i thought so, but how do you get that objective? As far as i know i have been everywhere and read everything so obviously i've missed something....

I'm playing on Hotshot. Sorry, I should have said that before. I don't think that Ice Axe is very obvious. It would make more sense if it went into inventory when you pick it up so you could tell it's not just a regular, of-no-particular-use item that you find lying around in most missions. Anyway, got the ice, now I have to figure out what to do with it.
