For panels, highlight them (you may need to bob around) then hit the USE key or button. Some panels are activated by hidden switches elsewhere in the room or level. Most secret panels will highlight, but some will not open unless you find the switch. For example, the secret passage from the Casino to the Guild den is activated by a torch inside, and a roulette wheel outside even though it highlights when you get a good look at it.
For the keys, bring them up in inventory and aim them at the highlighted lock and hit USE.
