I have run T2Fix_1.27b, minimal options, changed the audio settings as instructed here, started again. Got back to the factory Assembling Section area as fast as I could, heard Karras's speech, and the door has now opened. Thanks.

So Karras35 send me your save, Mabrien. I'm not Dromeder to know how to solve the issue and I'm aware that you restart mission and made door opened. But if you are curious, I managed to open this door. To do that I went to small room with desk and golden angel painting, very close to the place where you put Vibrant Stone. I went on the edge of window and made difficult jump to north platform with bronze gear above center door. From there I went to Break Room and to Security Room. After grabbing book, Karras spoke and after his speech door to Throne Room opened. I also grabbed Factory key and went there.
First of all small bot is still standing at the middle of hall instead of leaving it. And also turrets are active. Maybe it's because you scared him when you were attacking door. I don't know. So I was forced to strafe forward to avoid shots. I realized that cyborgs are awaken, so I strafe run to Throne Room elevator. When I approach it, I encountered massive lag, which only stopped when I arrived on top floor.
Another thing screwed in your save is Kernel, tracking cyborg. He doesn't move, he doesn't react to Garrett at all and he doesn't react to your attacks (he can't be put on ground by them either). So you kinda have easy mode.
So I'm just guessing that in your old run, scripts weren't loaded correctly. I hope you will not encounter any problems in your next run. Well, maybe except Kernel.

I have searched every part of the basement, either it's really really obvious and I am really really obtuse - or there's a bug with my copy. So if anyone could spoil me, I'd be very grateful, I've spent 3 hours going around in circles.

First check map of the basement. You are starting mission at "Entrance Here" location. Use control panel to open Air Shaft. Go all the way south to Elevator to the Roof. Double door are closed and unfrobbable. Switch to open elevator is right above door. So you need to go east to stairs leading to Elevator Controls. And you will appear above elevator door. Use lever to open door and use nearby switch to summon elevator. Go back to elevator and use inside switch to ride to the roof.
