Without creating a new object and adding the joints by hand, is there any way to force a Tweq Joints to rotate at a different position?

If the object doesn't have joints, you can't move them. If it does, you establish the base position using Shape > Joint positions. if you want to manipulate the existing joints outside of Tweq > Joints, then NVJointControl is the script for you.
